Jessica begins as the concubine to Duke Leto Atreides on Caladan before moving to Arrakis. Instructed by her mentor Reverend Mother Gaius Helen Mohiam to bear only daughters for the Bene Gesserit breeding program, Jessica instead intentionally conceived a son, Paul, out of love for Leto. This act created the Kwisatz Haderach, defying centuries of genetic planning. On Arrakis, after Duke Leto is killed by Baron Harkonnen and his traitorous doctor Wellington Yueh, Jessica and Paul escape into the desert. There, they utilize legends planted by the Missionaria Protectiva; Jessica casts herself as a Reverend Mother and Paul as the Lisan al'Gaib. She undergoes the spice agony ritual while pregnant, becoming a full Reverend Mother and giving birth to Alia, an Abomination who possesses other memories from birth. Later, in Children of Dune, she returns to Caladan and acts as a distant counselor to Emperor Paul. She eventually trains Prince Farad'n in the Bene Gesserit way after fleeing an assassination attempt by her possessed daughter, Alia.

The events surrounding Jessica's life are marked by intense conflict and transformation. In Dune, she is drugged, bound, and gagged by Harkonnen forces to prevent her from using her powers before being ordered for disposal in the desert. Relying on supplies left by Yueh, she and Paul find refuge with the Fremen. There, Jessica replaces a dying Reverend Mother after surviving the spice agony while pregnant, exposing her fetus to ancestral memories. Her daughter Alia is born as an Abomination, a full Reverend Mother from birth in a child's body. Years later, in Children of Dune, Jessica returns to inspect Paul's children, Leto II and Ghanima. Realizing Alia is fully possessed by ancestral personalities, she survives an assassination attempt and flees to Sietch Tabr with Stilgar. She is eventually found by Duncan Idaho, who kidnaps her on orders from the mysterious Preacher (suspected to be Paul) and takes her to Salusa Secundus. There, she trains Prince Farad'n before returning to Caladan, where she lives out her final 102 years.
